RTStable DMEM Media
with High Glucose, Sodium Bicarbonate
DMEM (Dulbecco’s Modified Eagle Medium) is a widely used basal medium designed to support the growth of a broad range of mammalian cell types. Cells routinely cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as established cell lines such as HeLa, HEK-293, Cos-7, and PC-12.
RTStable DMEM is specifically engineered for storage at room temperature while maintaining cellular proliferation rates and maximum cell densities comparable to conventional DMEM when supplemented with ≥10% fetal bovine serum (FBS).
This DMEM is modified as follows:
With: High Glucose, RTStable Glutamine, Phenol Red
Without: HEPES
Key modifications of RTStable DMEM include
- Optimization for room-temperature stability
- High glucose formulation with RTStable glutamine and phenol red

Using RTStable DMEM
DMEM differs from other basal media by containing approximately four-fold higher concentrations of amino acids and vitamins compared to the original Eagle’s Minimal Essential Medium. Although originally formulated with low glucose (1 g/L) and sodium pyruvate, DMEM is commonly used in higher-glucose formulations, with or without sodium pyruvate, depending on cellular requirements.
DMEM employs a sodium bicarbonate buffering system (3.7 g/L) and therefore requires a 5–10% CO₂ atmosphere to maintain physiological pH. The inclusion of RTStable glutamine minimizes toxic ammonia accumulation, thereby enhancing cell viability and supporting robust cell growth in a convenient, ready-to-use format. RTStable DMEM is protein-, lipid-, and growth factor-free and must be supplemented with ≥10% fetal bovine serum (FBS) prior to use.
