RTStable RPMI 1640 Media
RPMI 1640 Medium (Roswell Park Memorial Institute 1640) was originally developed for the cultivation of human leukemic cells in both suspension and monolayer cultures. It has since been shown to support the growth of a wide range of mammalian cell types, including HeLa, Jurkat, MCF-7, PC12, peripheral blood mononuclear cells (PBMCs), astrocytes, and various carcinoma cell lines. Multiple RPMI 1640 formulations are available to accommodate diverse cell culture applications.
RTStable RPMI 1640 Medium is engineered for room-temperature storage while maintaining strong cellular proliferation and achieving maximum cell densities comparable to those obtained with conventional RPMI 1640 Medium when supplemented with ≥10% fetal bovine serum (FBS).
RTStable RPMI 1640 Medium modifications include:

Using RTStable RPMI 1640 Medium
RPMI 1640 Medium differs from many other culture media due to the presence of the reducing agent glutathione and relatively high concentrations of vitamins. It contains biotin, vitamin B₁₂, and para-aminobenzoic acid (PABA), which are absent from Eagle’s Minimal Essential Medium and Dulbecco’s Modified Eagle Medium. Additionally, inositol and choline are present at significantly elevated levels.
RPMI 1640 Medium utilizes a sodium bicarbonate buffering system (2.0 g/L) and therefore requires a 5–10% CO₂ atmosphere to maintain physiological pH. The inclusion of RTStable Glutamine reduces toxic ammonia accumulation, enhancing cell viability and growth in a convenient, ready-to-use format. As the medium contains no proteins, lipids, or growth factors, supplementation with ≥10% fetal bovine serum (FBS) is required for optimal cell culture performance.
